ABSTRACT

Objective:To evaluate the clinical efficacy of Zhiwei pill for bile reflux gastritis (BRG) with symptoms of disharmony of liver and stomach, evaluate the safety after treatment by three-arm study, and investigate the regulatory action to gastrointestinal hormones. Method:One hundred and eighty patients were randomly divided into placebo group (60 cases), control group (60 cases) and observe group (60 cases) by random number table. Patients in three groups got lifestyle intervention of non-drug therapy. Patients in control group got hydrotalcite tablets 1-2 hours after meal or untimely bedtime or in case of stomach pain, 2 tablets/time, 3 times/day. Patients in placebo group got simulated medicine of Zhiwei pill 1 hour after meal, 6 g/time, 3 times/day. Patients in observe group got Zhiwei pill 1 hour after meal, 6 g/time, 3 times/day. The treatment continued for 4 weeks in all 3 groups. Before and after treatment, scores were graded for symptoms of disharmony of liver and stomach and patient reported outcome (PRO) scale for patients with chronic gastrointestinal diseases. Gastroscopy was taken to grade the scores of bile reflux and mucosa under gastroscope. Levels of gastrin (GAS), motilin (MTL), cholecystokinin (CCK), substance P (SP) and gastrointestinal hormones were detected. Efficacy for traditional Chinese medicine(TCM) syndrome and efficacy under gastroscope were compared, and safety after treatment was evaluated. Result:After treatment, the total scores of the main symptoms and 7 secondary symptoms of disharmony of liver and stomach in observe group were all lower than those in placebo group and control group (<italic>P</italic><0.01), and scores of symptoms of disharmony of liver and stomach and PRO were all lower than those in control group and placebo group (<italic>P</italic><0.01). After treatment, scores of degree of bile reflux and mucosa under gastroscope in observe group were all lower than those in placebo group and control group (<italic>P</italic><0.01). Level of CCK was lower than that in placebo group and control group (<italic>P</italic><0.01), while levels of MTL, GAS and SP were higher than those in placebo group and control group (<italic>P</italic><0.01). Total effective rate of TCM syndrome was 91.38%(53/58), higher than 71.93%(41/57) in control group and 34.62%(18/52) in placebo group. The total effective rate in gastroscopy was 93.10%(54/58), higher than 78.95%(45/57) in control group and 28.85%(15/52) in placebo group (<italic>P</italic><0.05 or <italic>P</italic><0.01). No adverse reactions related to Zhiwei pill were found. Conclusion:Zhiwei pill can improve bile reflux, promote the healing of mucosa under gastroscope, regulate gastrointestinal hormones, improve gastrointestinal motor function, significantly alleviate clinical symptoms, and improve quality of life, with better efficacy for TCM syndrome and efficacy in gastroscopy, and it is safe for clinical use.

ABSTRACT

<b>Objective::To study the mechanism of modified Taohe Chengqitang in preventing and treating diabetic gastroparesis by regulating relative genes and signaling pathways based on network pharmacology. <b>Method::Target genes of modified Taohe Chengqitang were obtained from Bioinformatics Analysis Tool for Molecular Mechanism of TCM (BATMAN-TCM) database, and target genes of diabetic gastroparesis were obtained from Comparative Toxicogenomics Database (CTD) database. The target genes of modified Taohe Chengqitang-diabetic gastroparesis intersection protein were obtained through the integration of two groups of genes. STRING was used to build the protein-protein interaction network and visualize the results. Cytospace software ClueGO, CluePedia plug-in were used for gene ontology (GO) analysis and enrichment analysis of KEGG pathway of modified Taohe Chengqitang-diabetic gastroparesis target genes intersection, and results were visualized. Finally, CTD database and literatures were used to obtain intersection genes in the treatment of diabetic gastroparesis. <b>Result::Among 621 target genes in modified Taohe Chengqitang, 25 were related to diabetic gastroparesis. By regulating expressions of MLNR, SST, PTGS1, HRH2, HTR3A, HTR4, HTR7, NOS3 and other intersection genes, Taohe Chengqitang could improve the gastrointestinal hormone levels, affected the combination of serotonin and its receptors, activated adenylate cyclase (AC), and induced cyclic adenosine monophosphate (cAMP) and protein kinase A (PKA), so as to activiate AC/cAMP/PKA signaling pathway, regulate Ca<sup>2+</sup> /K<sup>+</sup> channel, control ion balance, promote the adaptability of gastric smooth muscle, and contract gastric smooth muscle to regulate gastric volume. At the same time, gastric acid secretion was improved to protect gastric mucosa, which may help improve vasoconstriction and hemodynamics. <b>Conclusion::Based on the network pharmacology, modified Taohe Chengqitang has multiple mechanisms in the prevention and treatment of diabetic gastroparesis. This study explored relevant signaling pathways, advantages and research directions of modified Taohe Chengqitang in treatment of diabetic gastroparesis.

ABSTRACT

Objective To provide evidence for TCM treatment according to syndrome differentiation by studying the correlation between TCM syndromes and serum gastrin (GAS), motilin (MTL), cholecystokinin (CCK) and vasoactive intestinal peptide (VIP) levels of reflux esophagitis (RE) patients. Methods A total of 120 eligible RE patients from Dongzhimen Hospital were differentiated with TCM syndromes, and blood samples were taken to test for GAS, MTL, CCK and VIP levels. Differences between the different TCM syndromes in terms of serum gastrointestinal hormones levels were analysed. Results The male patients had the most distribution of stagnation of heat in the liver and stomach syndrome, but the female patients had the most distribution of disharmony of the liver and stomach syndrome (χ2=27.994, Ps<0.001). The Grades A, B and C were mainly associated with the obstruction of phlegm syndrome, disharmony of the liver and stomach syndrome, and stagnation of heat in the liver and stomach syndrome respectively (χ2=9.951, P=0.007). For patients with the syndrome of stagnation of heat in the liver and stomach, the serum GAS level was significantly lower than the syndrome of disharmony of the liver and stomach, the syndrome of obstruction of phlegm, the syndrome of deficiency of the center and upward flow of qi, and mixture of cold and heat syndrome (P<0.01 or P<0.05). For patients with mixture of cold and heat and patients with deficiency of qi and stagnation of blood, there was a significant raise in serum CCK level, compared to the patients with stagnation of heat in the liver and stomach, patients with disharmony of the liver and stomach, or patients with deficiency of the center and upward flow of qi (P<0.01). Compared with the patients with disharmony of the liver and stomach and patients with the syndrome of obstruction of phlegm, the serum CCK level of patients with the syndrome of deficiency of the center and upward flow of qi rose significantly (P<0.01). Compared with the patients with the syndrome of stagnation of heat in the liver and stomach, the serum MTL levels of patients with obstruction of phlegm and patients with mixture of cold and heat rose significantly (P<0.01). There was a significant raise in serum MTL level of patients with the syndrome of mixture of cold and heat compared to patients with disharmony of the liver and stomach and patients with deficiency of the center and upward flow of qi (P<0.05). Conclusions The RE patients with mixture of cold and heat, and patients with deficiency of qi and stagnation of blood, and patients with deficiency of the center and upward flow of qi may be related to the rise in CCK levels. The patients with the syndrome of stagnation of heat in the liver and stomach are related to the decrease in GAS and MTL levels.

ABSTRACT

Objective To investigate the clinical efficacy of trimebutine combined with Xiangsha Pingwei in the treatment of functional dyspepsia and its effect on gastrointestinal hormone.Methods One hundred and twenty cases of functional dyspepsia were randomly divided into the control group and observation group,60 cases in each group.The control group was treated with trimebutine 0.2 g,3 times/d and the observation group was treated with trimebutine 0.2 g,3 times/d and Xiangsha Pingwei 10 g,2 times/d.Both groups were treated for four weeks,a course of treatment.Clinical efficacy and the influence on gastrointestinal hormone levels were compared between the two groups.Results The symptoms scores in both groups all decreased significantly after treatment.While the postprandial fullness scores,early satiety scores and epigastric pain scores in the observation group were significantly lower than those in the control group (0.62±0.12 vs.1.85±0.25,0.56±0.14 vs.1.46±0.21,0.48±0.15 vs.1.32±0.12,t=34.36,27.62,33.87,P<0.05).The total effective rate in the observation group was significantly higher than that in the control group (90.00% vs.73.33%,χ2=5.57,P<0.05).MOT and GAS levels in both groups increased significantly.While MOT and GAS levels in the observation group were significantly higher than those in the control group (MOT: (355.2±78.1) ng/L vs.(288.7±82.4) ng/L;GAS:(96.5±17.6) ng/L vs.(88.4±16.0) ng/L,t=4.54,2.64,P<0.05).The somatostatin (SS) level in both groups decreased significantly.While the SS level in the observation group was significantly lower than that in the control group ((41.1±11.1) ng/L vs.(48.9±12.9) ng/L,t=3.55,P<0.05).Conclusion Trimebutine combined with Xiangsha Pingwei can significantly improve the clinical symptoms and gastrointestinal hormone in patients with functional dyspepsia.

ABSTRACT

Objective To observe the efficacy of laparoscopic surgery combined with sodium hyaluronate in the treatment of adhesive intestinal obstruction and its influence on disease-related indexes.Methods 180 cases of patients with adhesive ileus from September 2010 to April 2014 were selected and randomly divided into control group and observation group two groups, 90 cases in each group.The control group was treated with laparotomy and the observation group was treated by laparoscopic surgery combined with sodium hyaluronate.Then the operation related indexes such as operation time, intraoperative bleeding volume, postoperative gastrointestinal function recovery time, hospitalization time and complications of two groups were compared.And the serum levels of gastrointestinal hormone such as GAS, MTL, CCK and SS and inflammatory cytokines such as hs-CRP, IL-2, IL-6, IL-10 and TNF-αof two groups before and after treatment were detected and compared.Results The intraoperative bleeding volume of observation group was less than control group, postoperative gastrointestinal function recovery time and hospitalization time were shorter than control group, the incidence of complications was lower than control group, the difference was statistically significant (P <0.05).1, 4 weeks postoperative, the serum levels of gastrointestinal hormone such as GAS, MTL and SS of observation group were higher than control group, while the CCK was lower than control group, the difference was statistically significant (P<0.05).1, 4 weeks postoperative, the serum levels of inflammatory cytokines such as hs-CRP, IL-2, IL-6 and TNF-αof observation group were lower than control group, the difference was statistically significant (P<0.05).Conclusion Compared with the traditional laparotomy, laparoscopy combined with sodium hyaluronate has the advantages of small effect on patients with digestive function and postoperative inflammatory reaction, thus helps to shorten the recovery time of patients after surgery and inhibition the occurrence of complications such as recurrent intestinal obstruction.

ABSTRACT

Objective To investigate the effect of jejunal infusion of amino acids on secretion of gastrointestinal hormone in healthy dogs.Methods Six healthy adult dogs were treated with jejunal fistulas and femoral vein intubation.Twenty-four hours after the operation,solution of 8 different amino acid monomers (experimental group) or normal saline (control group) were infused into the jejunum of the dogs every 24hours.The levels of cholecystokinin (CCK),motilin,and gastrin in the peripheral plasma were measured using radioimmunoassay at the start of infusion (0 minute),and 30,60,90,and 120 minutes after infusion.Results Compared with the control group,the serum CCK level in the phenylalanine group was significantly higher 30 and 60 minutes after infusion [(1.25 ±0.19) ng/L vs.(0.66 ±0.14) ng/L,(1.23 ±0.12) ng/L vs.(0.80 ± 0.03) ng/L,both P < 0.01],while that in the tryptophan group was significantly higher 30 minutes after infusion [(1.08 ±0.26) ng/L vs.(0.66 ±0.14) rig/L,P <0.01].The other measurement results showed no statistically significant differences.Conclusions Jejunal infusion of phenylalanine or tryptophan may stimulate the secretion of gastrointestinal hormone to some extent.Aromatic amino acids (phenylalanine and tryptophan) is more potent in triggering the release of CCK than aliphatic (leucine,isoleucine,and methionine) and charged amino acids (aspartic acid,arginine,and glutamate).The mechanism may be related to the properties of the amino acids.

ABSTRACT

This article was aimed to study the effect of Baohe granules for patients with chronic heart failure (CHF) of traditional Chinese medicine (TCM) syndrome and the effect of gastrointestinal hormone secretion. Patients with CHF were used as the object of study. A total of 80 selected patients were divided into 2 groups. The control group was treated with western medicine anti-heart failure therapy. The treatment group was treated with the combination of Baohe granules. All patients were compared in aspects of TCM efficacy, symptom score and serum gastrin, motilin secretion after two-week medication. The results showed that both treatments can significantly improve the TCM syn-drome and symptom total score of CHF patients. And the effect of the treatment group with Baohe granules was bet-ter. The TCM single symptom integral was also obviously improved in the treatment group combined with Baohe granules, which included heart palpitations, shortness of breath, abdominal distension, and loss of appetite. The GAS and MTL levels of CHF patients were significantly increased in the treatment group. It was concluded that the treat-ment combined with Baohe granules can improve CHF patients with TCM syndromes, symptoms and gastrointestinal hormone secretion. Thus, it contributed to the stability of the disease condition. It can slow the disease progression and improve prognosis. So it is worth using widely in the clinical practice.

Functional dyspepsia is a common functional gastrointestinal disease , however , the cause of functional dyspepsia has not been fully elucidated .Ghrelin is braingut petide , secreted by major endocrine cells of the stomach ( x/A like cells ) .Its recep-tor is widely distributed in the body .It has many kinds of biological effects , such as regulating growth hormone secretion , feeding and energy balance , affecting neuroendocrine and gastrointestinal function and so on .A growing number of studies have shown that Ghrelin in FD has a positive role in increasing the improvement of food intake and generating hunger , promoting gastric emptying .Ghrelin is a new hot spot in the research of FD .

ABSTRACT

Objective: To explore the possible pathogenesis for functional dyspepsia (FD) patients with spleen deficiency and predominant dampness and investigate the intervention by Huoxiang Zhengqi Soft Capsule (HZSC). Methods: One hundred FD patients with spleen deficiency and predominant dampness were divided in two groups: the control and treatment groups. The patients in the treatment group were given HZSC+starch tablets and those in the control group were given Domperidone+simulated agent for treatment. After 28 d, the indexes of the symptom scores before and after the treatment, the efficacy judgement standard, gastric emptying, motilin (MTL), gastrin (GAS), nitric oxide (NO), and 5-hydroxytryptamine (5-HT) were observed and compared with the positive and healthy controls. Results: After the treatment by HZSC, the scores of epigastric fullness, early satiety, mouth sticky, nausea, vomiting, and defecate for patients were obviously decreased (P0.05). In the Domperidone treatment group there was no difference in the scores of epigastric fullness, early satiety, mouth sticky, nausea, vomiting, and defecate for patients (P>0.05). HZSC showed the significantly promoting effects on the gastric emptying of FD patients (P0.05). HZSC could increase MTL content obviously (P<0.05). Conclusion: HZSC could ameliorate the clinical symptoms in FD patients with spleen deficiency and predominant dampness, improve the gastric emptying, and regulate the gastrointestinal hormone levels.

ABSTRACT

Objective To investigate the therapeutic effects and mechanism of biliopancreatic diversion (BPD) surgery on type 2 diabetes mellitus(T2DM) in GK rats.Methods 16 GK rats were randomly divided into 2 groups:BPD surgery group included 10 rats undergoing BPD surgery,sham-BPD group included 6 rats undergoing a sham operation.Fasting plasma glucose,insulin,glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide(GIP)were detected one week before BPD surgery and the 1st week,4th week,10th week,26th week after BPD surgery.Oral glucose tolerance test(OGTT) and insulin tolerance test(ITT) were done in the 10th week after BPD surgery.Results There was no statistical difference in fasting plasma glucose,insulin,plasma GLP-1 or GIP between the 2 groups before surgery.Plasma glucose had significant reduction in BPD group compared to that in the sham group(P <0.05) and insulin level had no significant difference between the 2 groups.Rats in BPD group had significant improvement in glucose tolerance and insulin sensitivity compared to those in the sham group.Serum level of GLP-1 was significantly elevated in BPD group compared to that before surgery (P =0.0337 at the 1st week after surgery; P =0.0002 at the 4th week after surgery,P < 0.0001 at the 10th week after surgery,P <0.0001 at the 26th week after surgery) and that in sham-BPD group(P =0.0354 at the 1st week after surgery,P =0.0032 at the 4th week after surgery,P =0.0001 at the 10th week after surgery,P <0.0001 at the 26th week).Serum level of GIP was significantly lowered in BPD group compared to that before surgery(P =0.0189 at the 1st week after surgery; P =0.0007 at the 4th week after surgery,P =0.0003 at the 10th week after surgery,P <0.0001 at the 26th week after surgery) and that in sham-BPD group(P =0.0089 at the 1st week after surgery,P =0.0002 at the 4th week after surgery,P =0.0006 at the 10th week after surgery,P <0.0001 at the 26th week after surgery).The difference had statistical significance (P <0.05).Conclusion BPD surgery can significantly reduce fasting plasma glucose,improve glucose tolerance and insulin sensitivity.The change of serum levels of GLP-1 and GIP may play the major role in BPD treatment of diabetes mellitus.

ABSTRACT

Objective Through observing the clinical effect and the changes of gastrointestinal hormones caused by the treatment of different moxibustion time of the superficial gastritis patients due to the spleen and stomach weakness,to investigate the regulation of the dose-effect relation and the adjustment function on gastrointestinal hormone of serum of patients caused by the warming and nourish effect with moxibustion.Methods Eighty-four superficial gastritis Patients due to the spleen and stomach weakness were divided into group 1(treated by warming moxibustion for twenty minutes)(n=28),group 2(treated by warming moxibustion for forty minutes)(n=28),and drug(n=28)groups.The changes of the content of the Prostaglandin E2(PGE2)、Somatostatin(SS)and Epidermal growth factor(EGF)were observed before and after treatment between 3 groups and the clinical effect in different time.Results ①All 3 groups were compared after treatment,their clinical effect had no significant difference(P>0.05):②Compared with pre-treatment,the superficial gastritis symptom score had a very significant difference after the treatment(3.07±1.54)、(3.11±1.40)、(3.79±2.25)and during the 1 month follow-up(2.25±1.32)、(2.57±1.10)、(4.11±2.48),(P<0.01);③After treatment,the content of the PGE2 of the serum was increased obviously,compared with pre-treatment.There was a significant difference in each group(33.751±1.267)pg/ml、(33.774±8.583)pg/ml、(32.583±8.259)pg/ml,(P<0.05);After warming moxibustion for forty minutes,the content of the EGF of the serum was increased obviously,compared with pre-treatment,showing a significant difference(1.331±0.823)pg/ml,(P<0.05).Conclusion ①All of the three treatment methods had significant curative effect,and the curative effect had no significant difference among these 3 groups.But the moxibustion groups were markedly higher than the drug group in long-term result;②The moxibustion had established adjustment function on gastrointestinal hormone of the superficial gastritis patients due to the spleen and stomach weakness,which suggested that the gastrointestinal hormone may participate in its onset and the process of pathology and physiology.

ObjectiveTo investigate the effect of Qizil guliqent on gastrointestinal hormone in normal rats.MethodsThe rats in experimental groups were administered with Qizil guliqent(4g/kg,6g/kg,8g/kg ),madinglin (0.0027g/kg),and the rats in normal control group were given 0.5% CMC-Na for twenty days.The method ELISA was used to detect the content of gastric motility(MTL),vasoactive intestinal peptide(VIP) and Gastrin(GAS) in serum of normal rats.ResultsIn comparison with normal control group,VIP increased in Qizil guliqent group(4g/kg,6g/kg) (P <0.05 ~P < 0.01 );but MTL,GAS were not significantly changed in this group( P > 0.05 ).MTL,GAS were increased in Qizil guliqent group ( 8g/kg ) and madinglin group (0.0027g/kg) ( P < 0.05 ) ; but VIP was not significantiy changed in this group(P > 0.05 ).ConclusionQizil guliqent could regulate gastrointestinal hormones in rats,and dose related.

ABSTRACT

Objective To investigate the effect and mechanism of probiotic products on alpha-naphthylisothiocyanate(ANIT) induced intrahepatic cholestasis of rats.Methods Seventy-two SD rats were randomly divided into 3 groups: control group(n=8),intoxication group(n=32) and intervention group(n=32).After 3 days adapted feeding,all rats received a single intragastric administration of ANIT(200 mg/kg) to induce acute intrahepatic cholestasis.Probiotics[4.2?108/(kg?d)] was given to intervention group 2 days before ANIT was administrated.After ANIT was given,biliary flow,alanine aminotransferase transaminase(ALT),total bilirubin(TB) and gastrointestinal hormone were recorded in every 48 h till 192 h.SPSS 12.0 software was used to analyze the data.Results 1.Forty-eight hours after ANIT was gavaged,the biliary flow of intoxication and intervention groups were lowered while ALT and TB were increased.But the above observation items of intoxication group were more significantly altered than those of intervention group.Then,they all recovered gradually.Those of intervention group returned to nearly normal levels 144 h after ANIT was gavaged while those of the intoxication group recover at 192 h.2.After ANIT was gavaged,MTL of intoxication and intervention groups was lowered while vasoactive intestinal peptide(VIP) increased.But the above items of intoxication group were more significantly altered than those of intervention group.Then,they all recovered gradually and the intervention group was much more early to returned to normal levels than the intoxication group at 192 h.Conclusions Probiotics can promote the bowel movement through increasing the secretion of gastrointestinal hormone,then relieve the cholestasis.

[Objective] To observe the regulatory effect of astragalosides (AST) on the binding capacity of gastrin receptor in gastric parietal cells (GPC) of rats with spleen deficiency. [Methods] SD rats were given gastric gavage of Radix et Rhizoma Rhei (Dahuang) decoction 10 g/kg to induce spleen deficiency. Gastric parietal cells were isolated and purified by Lewin gastric sac method and Percoll density gradient separation method. 2 mL suspension of GPC from rat models was incubated in vitro with AST 20 ?L at the concentrations of 10, 20 and 30 g/L respectively. Suspension of GPC from rat models incubated with phosphate buffered saline (PBS) served as the model group and suspension of GPC from the normal rats incubated with PBS as the normal group. The binding capacity of gastrin receptor in GPC was detected by radioligand receptor assay. ABSTRACT

Objective To study the role of plasma levels of gastrointestinal hormones and gallbladder kinetics in the pathogenesis of functional dyspepsia. Methods Thirty two patients with functional dyspepsia (FD) and 20 normal subjects as controls were studied. Gallbladder ejection fractions (GBEF) were determined with 99m Tc radionucleography, and plasma levels of motilin (MTL)、cholecystokinin (CCK)、 vasoactive intestinal peptide (VIP) and somatostatin (SS) were measured with radioimmunoassay. Results Both preprandial and postprandial GBEF and MTL levels were markedly lower in FD group than those in control group ( P 0.05), while the levels of postprandial CCK were significantly decreased in FD group ( P 0.05). Conclusion (1) Patients with FD might have a decreased level of preprandial and postprandial gallbladder emptying. The decrease of plasma levels of MTL、CCK and the elevation of VIP might be the cause of gallbladder slow emptying in FD. (2) Radionucleography is non invasive and safe. It can be used in the quantitative analysis and dynamic study of gallbladder and Oddi sphincter functions, and provide accurate, physiological results.

ABSTRACT

Objective To study the correlations between the TCM syndromes of gastrointestinal dyskinesis and the disorders of gastrointestinal hormone,electrogastrogram(EGG) and blood gas in patients with chronic obstructive pulmonary diseases(COPD) and cor pulmonale.Methods The 120 cases of COPD and cor pulmonale were diagnosed with TCM syndrome differentiation,and plasma motilin(MOT),vasoactive intestinal peptide(VIP),Gastrin(GAS),EGG and blood gas were anglyzed and compared with 26 healthy subjects.Results With the aggravation of COPD and cor pulmonale,the dyskinesis of stomach and intestine was getting worse,developed from Shi-excess to Xu-deficiency.In deficiency syndrome,basic electrorhythm decreased most obviously.In phlegm-damp in lung syndrome,MOT and GAS decreased most obviously.In lung-kidney deficiency syndrome and lung-spleen-kidney deficiency syndrome,VIP increased most obviously.In lung-spleen-kidney deficiency syndrome,anoxia and carbon dioxide retention were most serious but GAS increased.Conclusion All patients of COPD and cor pulmonale have dyskinesis of stomach-intestine to a certain degree,abnormal EGG and disordered gastrointestinal hormones.Severity of ventilatory disturbance,quantity of injured organs,and condition of anoxia due to gastrointestinal blood stasis are all correlated with the development of COPD and cor pulmonale.

ABSTRACT

Objective To detect normal rat jejunum epithelial cells to learn about the distribution of peptide YY(PYY)receptor and measured two important parameters of PYY receptor so as to explore the target that PYY binds to and display its physiological function.Methods Receptor radioligand assay was conducted by using 125I-PYY to detect normal rat jejunum epithelial cells.We also measured receptor dissociation constant(Kd),the affinity or the ability that the receptor could bind ligand;maximum binding capacity(Bmax),the concentration of binding site;and Hill coefficient.Results There were PYY receptors in normal rat jejunum epithelial cells.Dissociation constant(Kd)of PYY receptor was(386.69?129.95)pmol/L.Maximum binding capacity(Bmax)was(303.21?116.85)fmol/mg protein.Hill coefficient was 1.PYY receptor had high affinity.Conclusion There are distributions of PYY receptors in normal rat jejunum epithelial cells.Kd means that PYY receptor has high affinity.Bmax means that the number of receptors is limited and decides the characters of saturability.We think that PYY directly inhibits secretion by binding to PYY receptor in jejunum epithelial cells when PYY inhibiting jejunum secretion.
